can i say to my employee "I will kill you " when she refuses my order?

You can say it, but depending on the circumstances, you may be committing a crime by doing so.  In most cases, that sort of hyperbole is not a criminal matter, but in particular cases where the circumstances could lead the threatened party to believe and be frightened by the threat, it could violate criminal statutes.
