More details to this question:
I worked at a gas station. My manager was also my uncles girlfriend. I was fired. when i was fired i was told it was because inventory was short. i called my mother and she already knew. my manager called her and told her i had been stealing money from the register which is absolutly untrue. i come from a wealthy family and have no need to steal. Because my manager is also my uncle's girlfriend my whole family knows about this and now i have the reputation of being a thief.

It looks as if you do have a defamation case, but you will probably have to prove that your manager knew that what she said was false in order for you to win.  The amount you would win is also a big question.  This appears to be the type of defamation which is categorized as per se, meaning that you do not have to allege actual monetary damages in order to state a claim, but the amount you would win could be minimal.
